Of course some courses are more flat than others and spectators may be more plentiful at bigger races, but when it comes down to it you still have to run 26.2 miles
You can adapt your training to any course you want to...
If you are running a hilly course then run hills
If you are running the Towpath Marathon then run on the Towpath
The Towpath Marathon was my first marathon and I would suggest it as a great first time marathon

Subject: [towpath_marathon] Question about the course.
I'm another first time marathon runner considering running the Towpath
first. I was wondering if any experienced marathoner could compare
this run to one like the chicago or other bigger city marathon. I have
a feeling it might be harder finding the motivation if there's less
people around.
If someone might know the pros and cons of running this marathon to
others, I'd be very interested to hear about it.
Thanks in advance,
DJ
