No. Just remember 8.05(l) "the pitcher, after coming to a legal pitching position, removes one hand from the ball other than in an actual pitch, or in throwing to a base."
So once he is in the set or windup position, he cannot break his hands apart. However, it has been accepted to allow the pitcher to momentarily adjust the ball in his glove before coming set or assuming the windup position. This adjustment must be brief though. If he has his hands together long enough that you think he has come set or assumed the windup position, then it is a balk if he breaks his hands.

Also, I don't know why any pitcher would take his signs with the ball in his glove. In order to pick someone off, he now has to step, reach into his glove, take the ball out and throw. If it's in his hand, he just has to step and throw.

The number of coaches is determined at the game - not from the number of players on the roster.  If you have 12 eligible players at the game, then the team only gets a manager and a coach.  If the team has 13 or more eligible at the game, then they get one manager and two coaches.  Of course these must be coaches named on the affidavit (this could be a temporary situation also, as noted in the regulation below):

"Managers/Coaches in the Dugout - If a tournament team has thirteen (13) or more
eligible players in uniform at a game, then the maximum of three (3) adults who are named
on the affi davit (or authorized temporary replacements as noted on the affi davit) will be
permitted to act as manager/coaches for that game. However, if a tournament team has
twelve (12) or fewer eligible players in uniform at a game, then a maximum of two (2) adults
must be named at the start of the game as manager and coach. The two named adults
must be listed on the affi davit, or must be authorized temporary replacements as noted on
the affi davit. If there is a third adult listed on the affi davit, that adult is not permitted to be in
the dugout or on the fi eld during that game.

"Replacement of Player, Manager or Coach
Any player, manager or coach listed on the Eligibility Affi davit who is unable to participate
because of injury, illness, vacation or other justifiable reason may be replaced by another
eligible person. If a player, manager or coach is replaced, that person may not be returned
to the Tournament Affi davit. Permanent replacements must be from the league’s regular
season teams and shall be recorded and approved by the District Administrator or Tournament
Director in the space provided on the Eligibility Affi davit. Exception: If a manager or coach
is unable to attend a game for a justifi able reason, a Tournament Director could approve a
temporary replacement. Temporary replacement of a manager or coach need not be entered
on the Eligibility Affi davit. A manager or coach who is ejected from a game may not be
replaced for the team’s next physically played game. (See Rule 4.07)"
