So once he is in the set or windup position, he cannot break his hands apart. However, it has been accepted to allow the pitcher to momentarily adjust the ball in his glove before coming set or assuming the windup position. This adjustment must be brief though. If he has his hands together long enough that you think he has come set or assumed the windup position, then it is a balk if he breaks his hands.
Also, I don't know why any pitcher would take his signs with the ball in his glove. In order to pick someone off, he now has to step, reach into his glove, take the ball out and throw. If it's in his hand, he just has to step and throw.
